What reconstitution means

Reconstitution is the process of dissolving a lyophilised (freeze-dried) powder in a diluent to produce a solution of known concentration for laboratory handling. The mass of material in the vial does not change; adding a measured volume of diluent simply converts that fixed mass into a liquid of defined concentration, expressed in milligrams per millilitre or micrograms per millilitre.

Choosing a diluent

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative, which suppresses microbial growth in a vial that will be accessed more than once. Sterile water contains no preservative and is suited to single-use laboratory preparations. The diluent choice does not affect the concentration maths — only the volume added does. Worked examples

  • A 5 mg vial with 2 mL of diluent gives 5 ÷ 2 = 2.5 mg/mL (2,500 mcg/mL).
  • A 10 mg vial with 2 mL of diluent gives 10 ÷ 2 = 5 mg/mL (5,000 mcg/mL).
  • A 2 mg vial with 1 mL of diluent gives 2 ÷ 1 = 2 mg/mL (2,000 mcg/mL).

Accuracy and verification

The calculated concentration assumes complete dissolution of the powder and accurate measurement of the diluent volume. Undissolved material, residue left on the vial wall, or an imprecisely measured volume will all shift the true concentration away from the calculated figure. Verify the stated vial mass and purity against the batch Certificate of Analysis supplied with the product before relying on the result.

Storage after reconstitution
